The present study used gene knockout mice to specifically test how deletion of p55 (TNFR1) in the TNF-alpha pathway might affect brain damage following ICH. Fourty-eight hours after ICH induction in mice, we observed an increase in TUNEL positive cells in the basal ganglia and an increase in pro-and active MMP-9 in p55(-/-) animals compared to WT. These data suggest an important protective role of p55 signaling of the TNF-alpha pathway against ICH induced brain damage. Ongoing study on the interplay of p55 with other signaling molecules and inflammatory factors will help us understand the molecular mechanisms of ICH pathology and identify potential targets for treatment of ICH.
