We applied optical techniques to visualize the subarachnoid space and cerebrospinal fluid (CSF) pathway by means of near infrared fluorescence of indocyanine green (ICG). ICG bound to lipoproteins was administered into the subarachnoid space (L5) of the adult rat. ICG fluorescence was excited by laser light (789 nm) and detected by a cooled-CCD camera with a bandpass filter (central wave-length 840 nm). After injection of ICG into the spinal subarachnoid space, ICG fluorescence was observed from the injection sire to the thoracic levels along the spinal subarachnoid space. In addition, one hour after the injection, ICG fluorescence extended to the cerebral subarachnoid space. The present results first demonstrate that In optical imaging technique is applicable to imaging of subarachnoid space and CSF pathway in vivo. In addition, ICG-lipoprotein provides a sensitive optical tracer for imaging biological structures outside blood vessels.
